I came home from work and noticed he's not swimming like he normally does so I took a closer look and on his right cheek, there's a single white spot. Can anyone tell me what this is?

Could be lymphocysts (spelling?)... have you checked your nitrates lately? Or is your tank new... possibly an ammonia spike? Lympho usually indicates poor water quality... I've experienced it in my QT tank before.
Its hard to tell from your pics and I'm not a disease expert by any means.
I had a pair for 2+ years sold it and the guy that had it added a flower pot for them to start breathing and the female beat the crap out of the male, sadly the male died, he gave me back the female to QT since his tank had ick. So sometimes things don't work out between them.
